SYLOID® FP Silica Pharmaceutical Excipients: Solving Flow and Caking Challenges

Technical Overview

Excellent flowability is critical for processing powdered active pharmaceutical ingredients (APIs) and excipients, as issues like agglomeration, caking, wall sticking, and electrostatic charging impair transportation, dosing, and blending. SYLOID® FP Silica, a series of synthetic amorphous silica excipients by W. R. Grace & Co.-Conn., effectively addresses these challenges with a small addition (0.2-2%).

The product line includes SYLOID® 244FP, AL-1FP (for EU), and 63FP (for US), all manufactured with strict quality control for high purity, full supply chain traceability, and compliance with USP-NF, JPE, and EP monographs. Its precisely porous, micronized structure enables unique intermolecular interactions, charge regulation, and capillary forces, delivering superior glidant and anti-caking performance.

Powder flow is influenced by raw material properties, interparticle interactions, and environmental factors (moisture, temperature, pressure). Hygroscopic products and incomplete drying cause agglomeration, but SYLOID® FP Silica’s high porosity adsorbs moisture to maintain formulation stability, outperforming other silica excipients. Elevated temperatures and prolonged pressure also induce caking, while its narrow particle size distribution enhances flow and blend uniformity, especially for direct compression.

For direct compression (valued for stability, cost-effectiveness, and simplicity), the two-step glidant mixing process uses AL-1FP/63FP in API pre-mixing to boost stability via hydrogen bonding with moisture, and 244FP in final mixing to prevent caking/segregation and provide essential processing properties. Washburn’s Equation helps predict powder wettability for formulation optimization.
