Pharmaceutical-Grade Silicon Dioxide in Tablet Glidant Applications: Driven by Both Compliance and Performance

With the upgrading of formulation technology, pharmaceutical excipient silicon dioxide has been widely applied in novel dosage forms such as orally disintegrating tablets and dispersible tablets, becoming a key material for pharmaceutical enterprises to improve product competitiveness.

As a multifunctional pharmaceutical excipient, pharmaceutical-grade silicon dioxide primarily serves as a glidant, anti-adherent, and disintegration promoter in tablet production. Its performance directly impacts the molding quality and bioavailability of pharmaceutical products. Pharmaceutical silicon dioxide complying with the standards of the Chinese Pharmacopoeia (ChP) features a purity of ≥99.5% and a particle size controlled between 1-10μm. The abundant silanol groups on its surface endow it with excellent adsorption and lubrication properties.
In the wet granulation tableting process, adding 0.1%-0.5% pharmaceutical-grade silicon dioxide can significantly reduce the friction between particles and between particles and die wall. This enhances the feeding and tablet ejection efficiency by 25% and improves tablet surface smoothness by 30%, effectively resolving production challenges such as sticking and capping. For tablets with high-potency ingredients, its uniform dispersion characteristic ensures the even distribution of active components, improving dosage accuracy. Meanwhile, it acts as an antistatic agent: by absorbing moisture in the air to form a hydrophilic film, it eliminates powder adhesion during fluidized bed granulation.
Compliance is one of the core competitiveness of pharmaceutical-grade silicon dioxide. Products must obtain FDA certification and meet stringent requirements including heavy metals ≤5ppm and microbial limit ≤100cfu/g. In commonly used tablets, it exhibits good compatibility with excipients such as microcrystalline cellulose and starch. Through co-grinding technology, it can refine the particle size of active pharmaceutical ingredients (APIs), shorten disintegration time, and enhance drug dissolution rate.
